Product Name | |
Generic Name | Recombinant Human IL-4 Protein |
Synonym | IL4,BCGF1,BSF1 |
Product Information | |
Construction | A DNA sequence encoding the human IL-4(GenBank: AAH70123.1 H25-S153)was expressed with a Fc-tag at the C-terminus. |
Expression Host | CHO cells |
QC Testing Purity | >90 % as determined by SDS-PAGE |
Activity | Determined by the dose-dependent stimulation of the proliferation of TF-1 cells.The expected ED50for this effect is ≤ 0.5 ng/ml. |
Endotoxin | < 0.1EU per 1 μg of the protein by the LAL method. |
Molecular Mass | The recombinant human IL-4 protein predicts a molecular mass of 15.4 kD. |
Formulation | Lyophilized from sterile PBS, pH 7.4. Normally 6 % mannitol are added as protectants before lyophilization. |
Stability & Storage | 24 months at 2℃ to 8℃ in lyophilized state. 6 months at -20℃ under sterile conditions after reconstitution. 12 months at -80℃ under sterile conditions after reconstitution. Recommend to aliquot the protein into smaller quantities after reconstituting with water for injection, normal saline or PBS, and keep the diluted concentration above 100μg/mL. Avoid repeated freeze-thaw cycles. |
Background | |
The role of IL-4 in the induction of monocytes into DC is to inhibit the excessive growth of macrophages, thereby guiding monocytes to differentiate towards DC. If IL-4 is not added to the culture system, monocytes will differentiate into macrophages. At the same time, IL-4 also has the ability to reduce the expression of CD14 molecules on the cell surface. The decrease in CD14 expression level is an important indicator of monocyte differentiation into DC. The combined action of GM-CSF and IL-4 can induce monocytes to differentiate into immature DCs (immature DCs), which have strong antigen uptake and processing abilities, but weak antigen presentation abilities. The cell surface moderately expresses MHCI class, II class molecules, and B7 family molecules (CD80, CD86, etc.), but does not express CD14. | |
