Product Name | |
Generic Name | Recombinant Human IL-1α Protein |
Synonym | L1A, IL-1α, IL1F1, IL1α |
Product Information | |
Protein sequence | A DNA sequence encoding the human IL-1α(P01583-1:S113-A271)was expressed with a His-tag at the C-terminus. |
Expression Host | HEK293 cells |
QC Testing Purity | > 90 % as determined by SDS-PAGE |
Activity | Determined by the level of interferon-γ secreted from induce human natural destruction lymphoma NK-92 cells. The expected ED 50 for this effect is ≤ 100 ng/ml. |
Endotoxin Level | < 0.1EU per 1 μg of the protein by the LAL method. |
Molecular Mass | The recombinant human IL-1αprotein predicts a molecular mass of 18.9 kD. |
Formulation | Lyophilized from sterile PBS, pH 7.4. Normally 6 % mannitol are added as protectants before lyophilization. |
Stability & Storage | 24 months at 2℃ to 8℃ in lyophilized state. 6 months at -20℃ under sterile conditions after reconstitution. 12 months at -80℃ under sterile conditions after reconstitution. Recommend to aliquot the protein into smaller quantities after reconstituting with water for injection, normal saline or PBS, and keep the diluted concentration above 100μg/mL. Avoid repeated freeze-thaw cycles. |
Background | |
Interleukin-1 (IL-1) is a peptide with a molecular weight of 17 kD and the presence of IL-1αand IL-1βtwo subtypes. IL-1αcontains 159 amino acids; IL-1β contains 153 amino acids; Both are encoded by different genes. Although its amino acid sequence has only 26% homology, IL-1αand IL-1βbind to the same cell surface receptors with the same affinity and exert the same biological effects. IL-1 receptors (IL-1R) are present on almost all nucleated cell surfaces, with varying numbers of IL-1R per cell, ranging from tens (such as t cells) to thousands (such as fibroblasts). There are two main types of IL-1R: one is IL-1r1, which has a longer peptide chain extending into the cytoplasm and plays a role in transmitting activation signals; The other is IL-1R2, which The peptide segment of the intracellular part is short, which can not effectively transmit signals. Instead, it releases the peptide chain of the extracellular part into extracellular fluid, and combines with IL-1 in a free form to play a feedback inhibitory role. GM-CSF, G-CSF, and IL-1 can all increase the expression level of IL-1R in cells, while TGF and corticosteroids can reduce the expression of IL-1R. | |
