Synonym | Long R3 IGF-1,Insulin-like Growth Factor-1,Somatamedin C,IGF-1A. |
Product Information | |
Protein sequence | A DNA sequence encoding the human IGF-1 LR3(PDB:3LRI_A)was expressed with a His-tag at the C-terminus. |
Expression Host | HEK293 cells |
QC Testing Purity | > 90 % as determined by SDS-PAGE |
Activity | Determined by the dose-dependent stimulation of the proliferation of MCF-7 cells. The expected ED 50 for this effect is ≤ 20 ng/ml. |
Endotoxin Level | < 0.1EU per 1 μg of the protein by the LAL method. |
Molecular Mass | The recombinant human IGF-1 LR3 protein consists of 89 amino acids and predicts a molecular mass of 9.9 kD. |
Formulation | Lyophilized from sterile PBS, pH 7.4. Normally 6 % mannitol are added as protectants before lyophilization. |
Stability & Storage | 24 months at 2℃ to 8℃ in lyophilized state. 6 months at -20℃ under sterile conditions after reconstitution. 12 months at -80℃ under sterile conditions after reconstitution. Recommend to aliquot the protein into smaller quantities after reconstituting with water for injection, normal saline or PBS, and keep the diluted concentration above 100μg/mL. Avoid repeated freeze-thaw cycles. |
Background | |
IGFS is a mitogenic peptide growth factor that can stimulate the proliferation and survival of various cells, including muscle, bone, and cartilage tissues. IGFS is mainly produced by the liver, although different tissues produce IGFS at different times. IGFS belongs to the insulin gene family and also contains insulin and relaxin. IGFS is similar in structure and function to insulin, but its growth promoting activity is significantly higher than insulin. The expression of IGF - II is influenced by placental prolactin, and the expression of IGF - I is regulated by growth hormone. IGF - I and IGF - II signals are transmitted through the tyrosine kinase type I receptor (IGF-IR), while IGF - II can also be transmitted through IGF - II/mannose 6-phosphate receptor signals. Mature IGFS is produced by protein hydrolysis of inactive precursor proteins containing N-terminal and C-terminal precursor regions. Recombinant human IGF - I and IGF - II are spherical proteins containing 70 and 67 amino acids and 3 intramolecular disulfide bonds, respectively. IGF-ILR3 is a recombinant analogue of human IGF - I, composed of a complete IGF-I sequence, with arginine substituted for the third glutamic acid and an N-terminal peptide extending 13 amino acids. The in vitro IGF-ILR 3, designed for higher biological efficacy, has an increased half-life and an aversion to binding to natural proteins in the body, making it an ideal choice for research and large-scale cultivation. | |
